Proanthocyanidine alleviates lung damage induced by nitrogen mustard

چکیده
Destructive properties of nitrogen mustard especially on respiratory system make it an effective chemical weapon with lack of an antidote. The aim of this study was to investigate the role of oxidative stress in mustard toxicity and the protective effect of proanthocyanidine. Rats were separated into three groups each containing fifteen rats. The three groups were control group which were exposed to none, nitrogen mustard group administered nitrogen mustard, and the last group administered nitrogen mustard and fed with proanthocyanidine containing diet (nitrogen mustard plus proanthocyanidine group). We demonstrated the harmful effects of nitrogen mustard over oxidative stress, and the therapeutic effects of proanthocyanidine by both histopathologically and biochemically using the oxidative stress parameters such as malondialdehyde, superoxide dismutase, catalase and glutathione peroxidase. Significant histopathologic lung lesions such as alveolar edema and alveolar congestion were observed in the nitrogen mustard group. Those lesions were less severe in the nitrogen mustard plus proanthocyanidine group. Proanthocyanidine treatment decreased malondialdehyde levels, which were initially elevated by nitrogen mustard administration, while superoxide dismutase, catalase and glutathione peroxidase activities remained similar to those of the nitrogen mustard only group. Our results revealed that oxidative stress has played a major role in mustard toxicity, and proanthocyanidine has had a substantial role in decreasing the toxic outcomes.

Overexpression of metallothionein in Chinese hamster ovary cells and its effect on nitrogen mustard-induced cytotoxicity: role of gene-specific damage and repair.
Overexpression of metallothionein in mammalian cells has been associated with protection from cytotoxic chemicals and acquired resistance of tumors to cytotoxic drugs. The mechanism of this effect, however, remains unclear. We have explored whether cytotoxicity of the bifunctional alkylating agent nitrogen mustard was correlated with the extent of DNA damage formation and repair in the metallot...

Sulfur mustard (SM), also known as mustard gas, has been the most widely used chemical weapon. The toxicity of SM as an incapacitating agent is of much greater importance than its ability to cause lethality. Acute toxicity of SM is related to reactive oxygen and nitrogen species, DNA damage, poly(ADP-ribose) polymerase activation and energy depletion within the affected cell. Therefore melatoni...

The response of two mustard (Brassica juncea L.) cultivars differing in photosynthetic capacity to different concentrations of hydrogen peroxide (H2O2) or nickel (Ni) was evaluated. Further, the effect of H2O2 on photosynthetic responses of the mustard cultivars grown with or without Ni stress was studied. Application of 50 μM H2O2 increased photosynthesis and growth more prominently in high ph...

Sulfur mustard (SM, bis- (2-chloroethyl) sulphide) is a chemical warfare agent that causes DNA alkylation, protein modification and membrane damage. SM can trigger several molecular pathways involved in inflammation and oxidative stress, which cause cell necrosis and apoptosis, and loss of cells integrity and function. Epigenetic regulation of gene expression is a growing research topic and is ...

Background and Aim: Sulfur mustard (Mustard gas) is the most famed chemical warfare agent that caused chronic lung damage. Oxidative stress is known as a major cause of mustard lung pathogenesis. This study aimed to introduce antioxidant tablets of Iranian N-acetyl-cysteine (NAC) as an effective treatment for chronic lung damage caused by mustard gas. Methods: This study was performed as a doub...
